Hiroshi Lockheimer (Senior VP of Android, Chromecast, Chrome OS at Google) has announced on Twitter that Google will start to roll out a newer version of Marshmallow as soon as next week for Nexus devices, containing several security bug fixes and of course the highly anticipated set of new emojis.
The update will also require a new system image meaning Google will release an OTA based off a future factory image (most likely going to be 6.0.1).
@rjcc new keyboard, new font, new code, so new system image. Nexus will update next week. OEMs decide their own update schedules.

For non-Nexus owners, you'll have to wait as OEMs are going to be responsible for updating their phones for support for this.
